Does it happen after their mother Ragyo has already been defeated (And Senketsu has perished leaving Ryko without a Kamui) or before that period while both girls (Unknowingly sisters) appear to be mortal enemies? I need to know to make my next move (I'll wait for you answer first before posting). Thanks!

What do you mean? Anyone can be the lead author. It really depends upon how the current story in the game is working out (It's a consensus among the "players" whether they want to continue with the story's current direction as the game goes along. If you want to be the lead author in your Kill la Kill game then I won't object (It's still too early in the game to determine that though).
